Novel combination therapy shuts down escape route, killing glioblastoma tumor cells

Glioblastoma, the most common and lethal form of brain tumor in adults, is challenging to treat because the tumors rapidly become resistant to therapy. As cancer researchers are learning more about the causes of tumor cell growth and drug resistance, they are discovering molecular pathways that might lead to new targeted therapies to potentially treat this deadly cancer.

Scientists at the Ludwig Institute for Cancer Research in San Diego worked collaboratively across the laboratories of Drs. Paul Mischel, Web Cavenee and Frank Furnari to investigate one such molecular pathway called the mammalian target of rapamycin or mTOR. This signaling pathway is hyperactivated in close to 90 percent of glioblastomas and plays a critical role in regulating tumor growth and survival. Therapies that inhibit mTOR signaling are under investigation as drug development targets, but results to date have been disappointing: mTOR inhibitors halt the growth but fail to kill the tumor cells.

A study published this week in the Proceedings of the National Academy of Sciences uncovers an unexpected but important molecular mechanism of mTOR inhibitor resistance and identifies a novel drug combination that reverses this resistance.

The story begins with a closer look at a gene-encoded protein called promyleocytic leukemia gene or PML. The study investigators explored the role of PML in causing resistance to mTOR inhibitor treatment. They found that when glioblastoma patients are treated with drugs that target the mTOR pathway, the levels of PML rise dramatically. Further, they showed that PML upregulation made the tumor cells resistant to mTOR inhibitors, and that if they suppressed the ability of the tumor cells to upregulate the PML protein, the tumor cells died in response to the mTOR inhibitor therapy.

"When we looked at cells in in vivo models and patients treated in the clinic, it became clear that the glioblastoma cells massively regulated PML enabling them to escape the effects of mTOR inhibitor therapy," reported senior author Paul Mischel, MD, Ludwig Institute member based at the University of California at San Diego.

"Our team hypothesized that if we could use a pharmacological approach to get rid of PML and combine it with an mTOR inhibitor, it could change the response from halting growth to cell death. The question was how?" added Mischel.

Previous research had shown that the use of low-dose arsenic could cause degradation of the PML protein in patients with leukemia. The team hypothesized that if arsenic could degrade PML, it may reverse resistance to mTOR inhibitors. The combination of mTOR and low-dose arsenic in mice indeed showed a synergistic effect, with massive tumor cell death along with very significant shrinkage of the tumor in mice with no ill side effects.

"Current therapy upregulates PML, turning off the mTOR signaling pathway. The tumor cells hide, waiting for the target signal to return," said Mischel. "When low-dose arsenic is added, not only does it stop the cell from returning, it shuts down the escape route killing the tumor cell."

These results present the first clinical evidence that mTOR inhibition promotes PML upregulation in mice and patients, and that it mediates drug resistance. The clinical relevance was confirmed when researchers looked at before- and after-treatment tissue samples from patients treated with mTOR inhibitors, confirming that PML goes up significantly in post treatment of mTOR inhibitors.

"These data suggest a new approach for potential treatment of glioblastoma," said Mischel. "We are moving forward to test that possibility in people."

Post-doctoral students Akio Iwanami and Beatrice Gini from the Mischel lab as well as Ciro Zanca from the Furnari/Cavenee lab, also contributed significantly to this paper.

TransAlta profit rises on lower maintenance costs

(Reuters) - Canadian power generation company TransAlta Corp reported a 58 percent rise in fourth-quarter profit, helped in part by lower maintenance costs.

Net earnings attributable to common shareholders rose to C$38 million ($37 million), or 15 Canadian cents per share, from C$24 million, or 11 Canadian cents per share, a year earlier.

Comparable earnings rose to 21 Canadian cents per share from 13 Canadian cents per share.

Revenue fell 6 percent to C$661 million.

Central Toronto Houses Hot

Seven of our top ten neighbourhoods are in central Toronto (west of DVP and east of Dufferin). In one half of the tale of two cities (the other being the gossamer that is Toronto's condoland), strong demand from urban families, coupled with a finite number of houses in central Toronto, means higher prices.

And in neighbourhoods offering detached houses or top schools, the appreciation is significant. The neighbourhoods on this list have appreciated 21% to 27%, well above the citywide appreciation rate of approximately 6%.

Inner Burbs Rise

Luxury Redefined

The Established Urban family drives demand in upper-end neighbourhoods like Allenby, Caribou Park, Rathnelly and Casa Loma on our list. They value the pedigree of the neighbourhood, larger houses and local schools, rather than their urban features per se, but do appreciate excellent subway access and top quality main streets.

Long-standing luxury neighbourhoods - Forest Hill, Bridle Path and Teddington Park - start on higher ground so it's rare to see them on "best appreciation" lists - so while Forest Hill makes our list, it's important to note that a handful of super luxury sales can skew the average.

Core Urban families value the "urban" in urban living - diversity, access to bike lanes and public transit and walkable main streets. Researchers marvel at world's deepest sea vents

KINGSTON, Jamaica (AP) ? Researchers using a remote-controlled submarine around the world's deepest known hydrothermal vents have been collecting samples from the sunless depths of the Caribbean Sea.

The expedition of Britain's National Oceanography Center is exploring areas where blazing hot, mineral-rich fluid gushes from volcanic chimneys on the seabed. The deepest areas are more than 3 miles (5 kilometers) below the sea's surface between the Cayman Islands and Jamaica.

Chief scientist Jon Copley says he believes laboratory analysis in coming weeks will reveal some new life forms that have evolved in the remote areas. In a Wednesday email from a research boat, Copley said these include bristle worms, a sea anemone and some small crustaceans.

The team was hailed in 2010 for discovering the deepest known volcanic vent field in the Cayman Trough.

Momentum grows for alternative phone system Tizen

BARCELONA, Spain (AP) -- The junior league of smartphone operating systems is getting more competitive. Phones from yet another contender ? Tizen ? will go on sale this year with a view to eventually competing with the industry leaders, Apple's iOS and Google's Android.

For now, Tizen will compete with another newcomer, Firefox OS, as well as Microsoft's Windows Phone and a revamped BlackBerry operating system.

Most of the impetus behind Tizen comes from cellphone carriers, which want a successful counterweight to the clout of Google and Apple. Samsung has become the world's largest maker of smartphones in large part through its embrace of Android.

Tizen has a powerful backer in Samsung Electronics Co. Spokesman Michael Lin confirmed Tuesday that the Korean company is ditching its own, homebrew operating system Bada in favor of Tizen. Samsung will also continue to make Android phones.

Yves Maitre, the executive in charge of handsets at Orange, France Telecom's wireless arm, said the carrier expects to launch Tizen phones in France this year and in developing countries next year. He spoke Tuesday to reporters and industry insiders at an event in Barcelona, Spain, on the sidelines of Mobile World Congress, the world's largest cellphone trade show.

Sprint Nextel Corp. is a member of the Tizen Association, but it hasn't said if it has any plans to bring Tizen handsets to the U.S. Other major backers include Intel Corp. and Huawei Technologies Ltd., China's largest phone maker.

Tizen phones will look and work much like Android phones, except that the familiar square app icons are round. By coincidence, that's the shape chosen for the Firefox OS, which was revealed in Barcelona on Sunday. Phones from that project are also expected this year.

Both Firefox and Tizen are "open source" projects, managed by non-profit associations. That means the software is freely available to customize, giving phone carriers control over how the software works on the phones they sell. By contrast, Apple maintains complete control over the workings of the iOS software in iPhones.

The idea to create an "open" phone operating system is not new. In fact, Android is an open-source project run by Google. However, one controlled by a non-profit has never been successful. Tizen is based on two failed attempts, MeeGo and LiMo. Frederic Dufal, the technical director of handsets at Orange, said The LiMo Foundation, the most broad-based attempt, was set up in a way that made it very hard for the interested parties to reach consensus. The nitty-gritty of getting Tizen to work is handled by the independent Linux Foundation instead, which has a history of shepherding the creation of effective software.

"We've learned from the mistakes of the past," Dufal said.

The Tizen Association said the phones will launch with a store with thousands of applications.

PHILADELPHIA (AP) ? Beer lovers across the U.S. have accused Anheuser-Busch of watering down its Budweiser, Michelob and other brands, in class-action suits seeking millions in damages.

The suits, filed in Pennsylvania, California and other states, claim consumers have been cheated out of the alcohol content stated on labels. Budweiser and Michelob each boast of being 5 percent alcohol, while some "light" versions are said to be just over 4 percent.

The lawsuits are based on information from former employees at the company's 13 U.S. breweries, some in high-level plant positions, according to lead lawyer Josh Boxer of San Rafael, Calif.

"Our information comes from former employees at Anheuser-Busch, who have informed us that as a matter of corporate practice, all of their products mentioned (in the lawsuit) are watered down," Boxer said. "It's a simple cost-saving measure, and it's very significant."

The excess water is added just before bottling and cuts the stated alcohol content by 3 percent to 8 percent, he said.

Anheuser-Busch InBev called the claims "groundless" and said its beers fully comply with labeling laws.

"Our beers are in full compliance with all alcohol labeling laws. We proudly adhere to the highest standards in brewing our beers, which have made them the best-selling in the U.S. and the world," Peter Kraemer, vice president of brewing and supply, said in a statement.

The suit involves 10 Anheuser-Busch products: Budweiser, Bud Ice, Bud Light Platinum, Michelob, Michelob Ultra, Hurricane High Gravity Lager, King Cobra, Busch Ice, Natural Ice and Bud Light Lime.

Anheuser-Busch, based in St. Louis, Mo., merged with InBev in 2008 to form the world's largest alcohol producer, headquartered in Belgium. In 2011, the company produced 10 billion gallons of malt beverages, 3 billion of them in the U.S., and reported $22 billion in profits from that category, the lawsuit said.

According to the lawsuit, the company has sophisticated equipment that measures the alcohol content throughout the brewing process and is accurate to within one-hundredth of a percent. But after the merger, the company increasingly chose to dilute its popular brands of beer, the lawsuit alleged.

"Following the merger, AB vigorously accelerated the deceptive practices described below, sacrificing the quality products once produced by Anheuser-Busch in order to reduce costs," said the lead lawsuit, filed Friday in federal court in San Francisco on behalf of consumers in the lower 48 states.

Companion suits are being filed this week in Pennsylvania, New Jersey and elsewhere. Each seeks at least $5 million in damages.

The named Pennsylvania plaintiffs, Thomas and Gerald Greenberg of Ambler, said they buy six cases of the affected Anheuser-Busch products a month. They did not immediately return a message Tuesday, and Boxer would not elaborate on their purchases except to say the consumer-protection suit does not involve retailers or bar owners.

One of the California plaintiffs, Nina Giampaoli of Sonoma County, said she bought a six-pack of Budweiser every week for the past four years.

"I think it's wrong for huge corporations to lie to their loyal customers ? I really feel cheated. No matter what the product is, people should be able to rely on the information companies put on their labels," Giampaoli said in a news release issued by Boxer's law firm.

Bloomberg News first reported Tuesday on the lawsuits.

In a telephone interview with The Associated Press, Boxer said he has evidence to corroborate the former employees' allegations, but stopped short of saying the beers had been independently tested.

"AB (Anheuser-Busch) never intends for the malt beverage to possess the amount of alcohol that is stated on the label. As a result, AB's customers are overcharged for watered-down beer and AB is unjustly enriched by the additional volume it can sell," the lawsuit said.

Ex-surgeon general C. Everett Koop dies

C. Everett Koop, who raised the profile of the surgeon general by riveting America's attention on the then-emerging disease known as AIDS and by railing against smoking, died Monday in New Hampshire. He was 96.

An assistant at Koop's Dartmouth institute, Susan Wills, said he died in Hanover, where he had a home. She didn't disclose the cause of his death.

Koop wielded the previously low-profile post of surgeon general as a bully pulpit for seven years during the Ronald Reagan and George H.W. Bush administrations.

An evangelical Christian, he shocked his conservative supporters when he endorsed condoms and sex education to stop the spread of AIDS.

He carried out a crusade to end smoking in the United States ? his goal had been to do so by 2000. A former pipe smoker, he said cigarettes were as addictive as heroin and cocaine.

Koop's impact was great, although the surgeon general has no real authority to set government policy. He described himself as "the health conscience of the country."

"My only influence was through moral suasion," Koop said just before leaving office in 1989.

By then, his Amish-style silver beard and white braided uniform were instantly recognizable.

Out of office, he switched to business suits and bow ties but continued to promote public health causes, from preventing childhood accidents to better training for doctors.

"I will use the written word, the spoken word and whatever I can in the electronic media to deliver health messages to this country as long as people will listen," he promised.

In 1996, he rapped Republican presidential hopeful Bob Dole for suggesting that tobacco is not invariably addictive, saying Dole's comments "either exposed his abysmal lack of knowledge of nicotine addiction or his blind support of the tobacco industry."

Although Koop eventually won wide respect with his blend of old-fashioned values, pragmatism and empathy, his nomination in 1981 met a wall of opposition from women's groups and liberal politicians.

Critics said Reagan selected Koop, a pediatric surgeon from Philadelphia, only because of his conservative views, especially his staunch opposition to abortion.

Foes noted that Koop traveled the country in 1979 and 1980 giving speeches that predicted a progression "from liberalized abortion to infanticide to passive euthanasia to active euthanasia, indeed to the very beginnings of the political climate that led to Auschwitz, Dachau and Belsen."

But Koop, a devout Presbyterian, was confirmed after he told a Senate panel he would not use the surgeon general's post to promote his religious ideology. He kept his word.

In 1986, he issued a frank report on AIDS, urging the use of condoms for "safe sex" and advocating sex education as early as third grade.

He also maneuvered around uncooperative Reagan administration officials in 1988 to send an educational AIDS pamphlet to more than 100 million U.S. households, the largest public health mailing ever done.

Koop personally opposed homosexuality and believed sex should be saved for marriage. But he insisted that Americans, especially young people, must not die because they were deprived of explicit information about how the HIV virus was transmitted.

He became a hero to AIDS activists, who chanted "Koop, Koop" at his appearances but booed other administration officials.

Koop further angered conservatives by refusing to issue a report requested by the Reagan White House, saying he could not find enough scientific evidence to determine whether abortion has harmful psychological effects on women.

Koop maintained his personal opposition to abortion, however. After he left office, he told medical students it violated their Hippocratic oath. In 2009, he wrote Senate Majority Leader Harry Reid urging that health care legislation include a provision to ensure doctors and medical students would not be forced to perform abortions. The letter briefly set off a security scare because it was hand delivered.

Koop served as chairman of the National Safe Kids Campaign and as an adviser to President Bill Clinton's health care reform plan.

At a congressional hearing in 2007, Koop spoke about political pressure on the surgeon general post. He said Reagan was pressed to fire him every day, but Reagan would not interfere.

Koop, worried that medicine had lost old-fashioned caring and personal relationships between doctors and patients, opened an institute at Dartmouth to teach medical students basic values and ethics.

He also was a part-owner of a short-lived venture, drkoop.com, to provide consumer health care information via the Internet.

Koop was born in New York's borough of Brooklyn, the only son of a Manhattan banker and the nephew of a doctor. He said by age 5 he knew he wanted to be a surgeon and at age 13 he practiced his skills on neighborhood cats.

He attended Dartmouth College, where he received the nickname Chick, short for "chicken Koop." It stuck for life.

Koop was by far the best-known surgeon general, and decades after he left the job he was still a recognized personality, colleagues recalled.

"I was walking down the street with him one time" about five years ago, recalled Dr. George Wohlreich, director of the College of Physicians of Philadelphia, a medical society with which Koop had longstanding ties. "People were yelling out, 'There goes Dr. Koop!' You'd have thought he was a rock star."

Dr. Joseph O'Donnell, an oncologist and professor at the Geisel School of Medicine, where the Koop Institute is located, said he shared Koop's desire to focus on disease prevention.

"When he decided he was going to come here I felt like I died and went to heaven," said Donnell, who is the senior scholar at the institute. "He was my hero, and we worked a lot together."

Koop received his medical degree at Cornell Medical College, choosing pediatric surgery because so few surgeons practiced it.

In 1938, Koop married Elizabeth Flanagan, the daughter of a Connecticut doctor. They had four children ? Allen, Norman, David and Elizabeth. David, their youngest son, was killed in a mountain-climbing accident when he was 20.

Koop's wife died in 2007, and he married Cora Hogue in 2010.

Koop was appointed surgeon-in-chief at Children's Hospital in Philadelphia and he also served as a professor at the University of Pennsylvania School of Medicine.

He pioneered surgery on newborns and successfully separated three sets of conjoined twins. He won national acclaim by reconstructing the chest of a baby born with the heart outside the body.

Although raised as a Baptist, he was drawn to a Presbyterian church near the hospital, where he developed an abiding faith. He began praying at the bedside of his young patients ? ignoring the snickers of some of his colleagues.

"It used to be said in World War II that there were no atheists in foxholes," he wrote in 1973. "I have found there are very few atheists among the parents of dying children.

"This is a time when religious faith can see a family through trying circumstances."

NEW YORK (AP) ? It looks like it's one and done at the Oscars for Seth MacFarlane.

The "Family Guy" creator was asked on his Twitter account whether he'd consider hosting the Academy Awards again and he replied: "No way. Lotta fun to have done it, though."

MacFarlane's edgy comedy proved a polarizing force on Sunday's Academy Awards, with jokes about domestic violence, women's bodies and Jews in Hollywood that offended some viewers. The Oscars did get their biggest audience in three years, however, with particular growth among young viewers.

MacFarlane's spokeswoman said Tuesday she had nothing to add to the tweet.

After quiet week, Jimmie Johnson makes some noise winning Daytona 500

There wasn't a whole lot of attention paid to the five-time NASCAR Sprint Cup champion last week. But when it counted most, Jimmie Johnson showed off his driving skills once again.

All the attention went to Danica Patrick and a handful of other drivers.

Not that it mattered Sunday, because look who pulled into Victory Lane.

Five-time is back. Not that he ever went away.

Johnson won his second Daytona 500 on Sunday, a year after he completed just one lap in the race and three months after falling short in his bid for a sixth Sprint Cup title. That so-called drought had made him something of a no-name during Speedweeks.

"In my mind, I didn't feel like I was under the radar," he said. "I felt like we were working hard to put the best product on the track. I guess I was quiet in the overall spectrum of things from the media side. I think people in the garage, people knew we were sitting on a lot of speed and had a very good race car."

But in winning the biggest race of the year, the No. 48 team wasn't sending a message to the competitors.

"I don't think we went anywhere; anybody in the garage area, they're wise to all that," Johnson said.

Johnson's win came on the same day that Patrick, who became the first woman in history to start a Sprint Cup race from the pole, again made history as the first woman to lead laps in the Daytona 500.

She ran inside the top 10 almost the entire race, kept pace with the field and never panicked on the track.

Her only mistakes were on pit road, where she got beat on the race back to the track, and on the final lap, when she was running third but got snookered by the veterans and faded to eighth. That's going to stick with Patrick for some time.

"I would imagine pretty much anyone would be kicking themselves about what they coulda, shoulda have done to give themselves an opportunity to win," she said. "I think that's what I was feeling today, was uncertainty as to how I was going to accomplish that."

There were several multicar crashes, but no one was hurt and none of them approached the magnitude of the wreck that injured more than two dozen fans in the grandstand at the end of the second-tier Nationwide Series race on the same track a day earlier. Daytona International Speedway workers were up until 2 a.m repairing the fence that was damaged in the accident, and track officials offered Sunday morning to move any fans who felt uneasy sitting close to the track.

Several drivers said the accident and concern for the fans stuck with them overnight and into Sunday morning, and Johnson was quick to send his thoughts from Victory Lane.

"I just want to give a big shout-out to all the fans, and I also want to send my thoughts and prayers out to everybody that was injured in the grandstands," Johnson said.

ASUS (2357) on Monday announced its next-generation PadFone during a press event at Mobile World Congress in Barcelona. The PadFone Infinity, which is a smartphone that can dock into a tablet, is equipped with a 5-inch 1080p display, a 1.7GHz quad-core Snapdragon 600 processor and a 13-megapixel rear camera. The device also includes 2GB of RAM, NFC, LTE, up to 64GB of internal storage, a 2,400 mAh battery and Android 4.2.

The Pad portion of the phone, which ASUS calls the Infinity Station, features a 10.1-inch full HD 1,920 x 1,200-pixel display and is capable of increasing the smartphone?s battery life to 40 hours or fully recharging the docked PadFone up to three times.

The PadFone Infinity will be offered in the U.K. for ?799 for the phone and an additional ?199 for the tablet dock. Like earlier models, the PadFone will not be available in the United States.

Protest votes increase uncertainty in Italy election

ROME (Reuters) - Italians voted for a second and final day in a general election on Monday with a surge in protest votes increasing the risk of an unstable outcome that could undermine Europe's efforts to end its three-year debt crisis.

Opinion polls give the centre-left coalition led by former Industry Minister Pier Luigi Bersani a narrow lead but the race has been thrown wide open by the prospect of protest votes against austerity and corporate and political scandals.

"I'm sick of the scandals and the stealing," said Paolo Gentile, a 49-year-old Rome lawyer who said he had voted for the 5-Star Movement, an anti-establishment protest group set to make a huge impact at its first general election.

"We need some young, new people in parliament, not the old parties that are totally discredited," he said.

Most of the voters interviewed outside polling stations by Reuters on Sunday and Monday expected the next government would quickly collapse, thwarting efforts to end an economic crisis.

"I'm very pessimistic, I don't think that whoever wins will last long or be able to solve the problems of this country," said Cristiano Reale, a 43 year-old salesman in Palermo, Sicily. He said he would vote for the far left Civil Revolution group.

A bitter campaign, fought largely over economic issues, has been closely watched by financial markets, nervous about the return of the kind of debt crisis that took the whole euro zone close to disaster and brought technocrat prime minister Mario Monti to office in 2011.

Italy is the third largest economy in the 17-member bloc and the prospect of political stalemate could cause dangerous market instability.

ANTI-EURO FORCES

"There are similarities between the Italian elections and last year's ones in Greece, in that pro-euro parties are losing ground in favor of populist forces," said Mizuho chief economist Riccardo Barbieri.

"An angry and confused public opinion does not see the benefits of fiscal austerity and does not trust established political parties."

Voting ends at 3 p.m. (1400 GMT), with the first exit polls due shortly afterwards. Projections based on the vote count will be issued through the afternoon and the final result should be known late on Monday or early Tuesday.

An extremely close Senate race is expected in several battleground regions and this could delay the final result.

Italian markets were sanguine on Monday morning, with both stocks and bonds showing little change.

Many analysts believe that whatever the result, the next government will be forced to stick to Italy's fiscal commitments because of the fear of a sudden spike in borrowing costs if markets take fright.

The election result is likely to be the most fragmented in decades, with the old left-right division disrupted by the rise of 5-Star, led by fiery Genoese comic Beppe Grillo, and by Monti's decision to run at the head of a centrist bloc.

"It will be a vote of protest, maybe of revolt," said Corriere della Sera, Italy's largest newspaper, on Monday.

It noted that for the first time the winning coalition is unlikely to get more than a third of the votes, making it harder to govern and likely opening weeks of complicated negotiations.

It is unclear how Grillo's rise will influence the result, with some pollsters saying it increases the chances of a clear win for the centre-left, led by Bersani's Democratic Party (PD), because 5-Star is taking votes mainly from Berlusconi.

After the first day of voting on Sunday, about 54 percent of voters had cast their ballots, a sharp fall on the level of 62.5 percent seen at the same stage in the last election in 2008.

If the trend continues on Monday it will confirm the disillusion of voters with a discredited political class.

Bad weather, including heavy snow in some areas, is also thought to have hampered the turnout in Italy's first post-war election to be held in winter.

This could favor the centre-left, whose voters tend to be more committed than those on the right, which has strong support among older people.

"Given the lower turnout, people are betting on a victory of the centre-left," said a Milan trader.

CALL TO ARMS

The 5-Star Movement, backed by a frustrated younger generation increasingly shut out of full-time jobs, could challenge former premier Silvio Berlusconi's People of Freedom (PDL) party as Italy's second largest political force.

"Come on, it isn't over yet," was Monday's front page headline in Il Giornale daily, owned by Berlusconi's brother, a call to arms to get out the vote.

The 76-year-old Berlusconi, has pledged sweeping tax cuts and echoed Grillo's attacks on Monti, Germany and the euro in an extraordinary media blitz that has halved the lead of the centre-left since the start of the year.

Support for Monti's centrist coalition meanwhile has faded after he led a lackluster campaign and he appears set to trail well behind the main parties.

Monti helped save Italy from a mounting debt crisis when he replaced a discredited Berlusconi in November 2011, but with the economy in its longest recession for 20 years analysts fear an electoral stalemate could delay vital moves to stimulate growth.

"I voted for the PD because a PD win is the only way to have a stable government and we need stability or we will end up like Greece," said Viola Rossi, an 80 year-old pensioner from Rome.

After drawing hundreds of thousands of supporters to its final campaign rally on Friday, Grillo has said he fears voting fraud to try to block a massive breakthrough, telling his supporters to wet the lead in the pencils they use to vote to prevent the crosses being rubbed out.

Whatever government emerges will inherit an economy that has been stagnant for much of the past two decades and problems ranging from record youth unemployment to a dysfunctional justice system and a bloated public sector.

If Bersani wins, it is far from clear that he will be able to control both houses of parliament and form a stable government capable of lasting a full five-year term.

Italy's electoral laws guarantee a strong majority in the lower house to the party or coalition that wins the biggest share of the overall national vote.

However the Senate, elected on a region-by-region basis, is more complicated and the result could turn on a handful of regions where results are too close to call, including Lombardy in the rich industrial north and the southern island of Sicily.

Many politicians and analysts believe Bersani and Monti will end up in an alliance after the vote, despite a number of spiky exchanges during the campaign and Monti's insistence that he will not join forces with Bersani's leftist allies.

As well as the national election, voters are also casting their ballots to elect new regional administrations in Lombardy, Lazio and Molise.
