Electrically spun fabric offers dual defense against pregnancy, HIV

The only way to protect against HIV and unintended pregnancy today is the condom. It's an effective technology, but not appropriate or popular in all situations.

A University of Washington team has developed a versatile platform to simultaneously offer contraception and prevent HIV. Electrically spun cloth with nanometer-sized fibers can dissolve to release drugs, providing a platform for cheap, discrete and reversible protection. The research was published this week in the Public Library of Science's open-access journal PLoS One. The Bill & Melinda Gates Foundation last month awarded the UW researchers almost $1 million to pursue the technology.

"Our dream is to create a product women can use to protect themselves from HIV infection and unintended pregnancy," said corresponding author Kim Woodrow, a UW assistant professor of bioengineering. "We have the drugs to do that. It's really about delivering them in a way that makes them more potent, and allows a woman to want to use it."

Electrospinning uses an electric field to catapult a charged fluid jet through air to create very fine, nanometer-scale fibers. The fibers can be manipulated to control the material's solubility, strength and even geometry. Because of this versatility, fibers may be better at delivering medicine than existing technologies such as gels, tablets or pills. No high temperatures are involved, so the method is suitable for heat-sensitive molecules. The fabric can also incorporate large molecules, such as proteins and antibodies, that are hard to deliver through other methods.

At a lab meeting last year, Woodrow presented the concept, and co-authors Emily Krogstad and Cameron Ball, both first-year graduate students, pursued the idea.

They first dissolved polymers approved by the Food and Drug Administration and antiretroviral drugs used to treat HIV to create a gooey solution that passes through a syringe. As the stream encounters the electric field it stretches to create thin fibers measuring 100 to several thousand nanometers that whip through the air and eventually stick to a collecting plate (one nanometer is about one 25-millionth of an inch). The final material is a stretchy fabric that can physically block sperm or release chemical contraceptives and antivirals.

"This method allows controlled release of multiple compounds," Ball said. "We were able to tune the fibers to have different release properties." One of the fabrics they made dissolves within minutes, potentially offering users immediate, discrete protection against unwanted pregnancy and sexually transmitted diseases.

Another dissolves gradually over a few days, providing an option for sustained delivery, more like the birth-control pill, to provide contraception and guard against HIV.

The fabric could incorporate many fibers to guard against many different sexually transmitted infections, or include more than one anti-HIV drug to protect against drug-resistant strains (and discourage drug-resistant strains from emerging). Mixed fibers could be designed to release drugs at different times to increase their potency, like the prime-boost method used in vaccines.

The electrospun cloth could be inserted directly in the body or be used as a coating on vaginal rings or other products.

Electrospinning has existed for decades, but it's only recently been automated to make it practical for applications such as filtration and tissue engineering.

This is the first study to use nanofibers for vaginal drug delivery.

While this technology is more discrete than a condom, and potentially more versatile than pills or plastic or rubber devices, researchers say there is no single right answer.

"At the time of sex, are people going to actually use it? That's where having multiple options really comes into play," Krogstad said. "Depending on cultural background and personal preferences, certain populations may differ in terms of what form of technology makes the most sense for them."

The team is focusing on places like Africa where HIV is most common, but the technology could be used in the U.S. or other countries to offer birth control while also preventing one or more sexually transmitted diseases.

The research to date was funded by the National Institutes of Health and the UW's Center for AIDS Research. The other co-author on the paper is Thanyanan Chaowanachan, a UW postdoctoral researcher and longtime HIV expert.

The team will use the new Gates Foundation grant to evaluate the versatility and feasibility of their system. The group will hire more research staff and buy an electrospinning machine to make butcher-paper sized sheets. The expanded team will spend a year testing combinations that deliver two antiretroviral drugs used to treat HIV and a hormonal contraceptive, and then six months scaling up production of the most promising materials.

NASA's Mars rover finds complex chemicals

Although NASA's Curiosity rover hasn't yet confirmed the detection of organic compounds on Mars, it's already seeing that the Red Planet's soil contains water and more complex chemicals ? including signs of an intriguing compound called perchlorate.

The first soil sample analysis from Curiosity's Sample Analysis at Mars lab, or SAM, was the leadoff topic today at the American Geophysical Union's fall meeting in San Francisco. The findings were eagerly awaited because of rumors that the Curiosity team was on the verge of announcing major findings?? and although NASA tamped down expectations, the scientists said they were overjoyed with?the first round of analysis.


"We really consider this a terrific milestone," Paul Mahaffy, a NASA researcher who is SAM's lead scientist, said at the AGU briefing.

Mahaffy said in a statement issued by NASA's Jet Propulsion Laboratory that "we have no definitive detection of Martian organics at this point, but we will keep looking in the diverse environments of Gale Crater."

Curiosity landed in Gale Crater on Aug. 5, and since then it's been studying Martian rocks, soil and atmosphere with a suite of 10 scientific instruments. Its two-year, $2.5 billion primary mission is aimed at determining whether conditions in the crater were ever conducive for microbial life.

SAM is a key tool for that mission, because it can cook Martian samples in a mini-oven and then analyze the gases that are given off to identify the compounds contained in the sample. Other instruments ? including the Chemistry and Mineralogy instrument, or CheMin ? were used as well to study the initial soil samples, collected over the past several weeks from a drift of windblown sand and dust called Rocknest.

This Oct. 31 view from Curiosity's Mars Hand Lens Imager, or MAHLI, shows two of the trenches made by the rover's 1.6-inch-wide (4-centimeter-wide) sampling scoop. The dust and sand from a dune known as Rocknest were fed into Curiosity's onboard labs for analysis.

NASA said CheMin found that the composition of the Rocknest samples was similar to that of soil analyzed by other Mars rovers such as Pathfinder, Spirit and Opportunity ? about half common volcanic minerals, and half non-crystalline minerals such as glass. SAM identified other ingredients in much lower concentrations, including water molecules that were apparently bound to the grains of sand and dust. Although the water wouldn't be enough to support any sort of life, the concentration was higher than expected.

SAM also identified a type of perchlorate, a compound that includes oxygen and chlorine. Perchlorate, which was also found by NASA's Phoenix Mars Lander in 2008, is considered a toxic substance and used as an ingredient in rocket fuel on Earth. But scientists say the compound could conceivably serve as an energy source for hardy microbes on Mars. Mahaffy said the particular type of compound detected by Curiosity appeared to be calcium perchlorate, but "we have to study that further."

Reactions with other chemicals in SAM's oven formed chlorinated methane compounds, which geologists consider organic chemicals because they contain carbon and hydrogen. Mahaffy said it was most likely that the chlorine came from a perchlorate-like compound in the soil. However, he said it wasn't yet clear whether the tiny amount of carbon in the compounds came from the Martian soil or was actually brought to Mars from Earth by Curiosity itself.

"We have to be very careful to make sure both the carbon and the chlorine are coming from Mars," he told reporters.

Caltech's John Grotzinger, the project scientist for Curiosity's mission, seconded that view. "We just simply don't know if they're indigenous to Mars or not," he said.

Grotzinger said the team would first have to confirm that the constituents of the organic compounds seen by SAM truly came from Mars. If the presence of organics is confirmed, then the scientists would have to look into whether they are merely part of the "background fall of cosmic material" onto the planet, or arose through chemical processes on Mars itself, he said.

It would take a step-by-step process to confirm the presence of truly Martian organic compounds, and reconstruct how those compounds were formed.?"Then you have ... to decide whether or not those formation pathways are abiotic, or maybe in the end biologic," Grotzinger said. "So you see there's a complicated decision pathway there, and we have to explore each one systematically."

Grotzinger cautioned that there would be no "hallelujah moment" in the search for organic chemicals on Mars.

A couple of weeks ago, he was quoted as saying that the data set from Curiosity would be "one for the history books." That led to speculation that an earth-shaking discovery could be revealed at the AGU meeting. NASA later said Grotzinger was referring to the two-year mission as a whole, rather than any specific findings to be announced in the near term.

Today, Grotzinger said his original comments were misunderstood.

"What I've learned from this is that you have to be careful about what you say, and even more careful about how you say it." he told reporters. "We're doing science at the speed of science [but] we live in a world that's sort of at the pace of Instagrams. The enthusiasm that we had, that I had, that our whole team has about what's going on here ... I think it was just misunderstood."

Update for 3:05 p.m. ET: After discussing this report with my friends in the newsroom as well as on Twitter, I've turned the headline around to emphasize the detection of chlorinated methane compounds, even though the origin of those compounds has not yet been confirmed. Is that going too far the other way?

Using genetic analyses, scientists have discovered that Northern European populations?including British, Scandinavians, French, and some Eastern Europeans?descend from a mixture of two very different ancestral populations, and one of these populations is related to Native Americans. This discovery helps fill gaps in scientific understanding of both Native American and Northern European ancestry, while providing an explanation for some genetic similarities among what would otherwise seem to be very divergent groups. This research was published in the November 2012 issue of the Genetics Society of America's journal GENETICS.

According to Nick Patterson, first author of the report, "There is a genetic link between the paleolithic population of Europe and modern Native Americans. The evidence is that the population that crossed the Bering Strait from Siberia into the Americas more than 15,000 years ago was likely related to the ancient population of Europe."

To make this discovery, Patterson worked with Harvard Medical School Professor of Genetics David Reich and other colleagues to study DNA diversity, and found that one of these ancestral populations was the first farming population of Europe, whose DNA lives on today in relatively unmixed form in Sardinians and the people of the Basque Country, and in at least the Druze population in the Middle East. The other ancestral population is likely to have been the initial hunter-gathering population of Europe. These two populations were very different when they met. Today the hunter-gathering ancestral population of Europe appears to have its closest affinity to people in far Northeastern Siberia and Native Americans.

The statistical tools for analyzing population mixture were developed by Patterson and presented in a systematic way in the report. These tools are the same ones used in previous discoveries showing that Indian populations are admixed between two highly diverged ancestral populations and showing that Neanderthals contributed one to four percent of the ancestry of present-day Europeans. In addition, the paper releases a major new dataset that characterizes genetic diversity in 934 samples from 53 diverse worldwide populations.

"The human genome holds numerous secrets. Not only does it unlock important clues to cure human disease, it also reveal clues to our prehistoric past," said Mark Johnston, Editor-in-Chief of the journal GENETICS. "This relationship between humans separated by the Atlantic Ocean reveals surprising features of the migration patterns of our ancestors, and reinforces the truth that all humans are closely related."

Russia urges North Korea not to launch rocket

MOSCOW (Reuters) - Russia urged North Korea on Monday not to go ahead with a plan for its second rocket launch of 2012, saying the launch would violate restrictions imposed by the U.N. Security Council.

"We urgently appeal to the government (of North Korea) to reconsider the decision to launch a rocket," the Russian Foreign Ministry said in a statement.

North Korea's state news agency on Saturday announced the decision to launch another space satellite and reportedly told neighbors it would take a similar path to a failed rocket launch in April.

Echoing its criticism of the April launch, Russia said Pyongyang had been warned not to ignore a U.N. Security Council resolution which "unambiguously prohibits (North Korea) from launching rockets using ballistic technology."

North Korea says its rockets are used to put satellites into orbit for peaceful purposes. The Russian statement said North Korea would be allowed only to exercise its right to peaceful activity in space if the U.N.-imposed restrictions were lifted.

Russia has often balanced criticism of Soviet-era client state North Korea's nuclear activities and missile launches with calls on other powers to refrain from belligerent actions against Pyongyang, which it says can be counterproductive.

Russia is a permanent member of the U.N. Security Council and is upset by any defiance of council resolutions. Past launches by Pyongyang have caused concern among Russians living near the country's border with North Korea.

The Email or the Phone Call: Which Comes First? | #b2bsales

Emails and phone calls are the two leading prospecting methods. So, have you ever wondered which should come first? Converting a prospect largely depends on the first contact. A prospect?s interest should pique during the first few seconds of the first communication. So, which is better: a phone call or an email?

The answer to our question isn?t black and white. In some situations, a phone call should always come first; in others, an email.

When Emails Come First
Email a prospect first when you don?t have enough information about them to know whether they?re a right fit. Don?t waste a lot of time calling cold leads. You can automate emails and insert content and forms into them that will ?help you understand what they?re interested in. An email campaign can increase brand awareness and nurture leads.

When Phone Calls Come First
Phone calls allow you to make a personal connection with a prospect, and convey your personality more than an email ever can. When you?ve done your research, and you know a prospect is a good fit, always call them before emailing them.

Phone calls are so effectual because they force action and conversation. Emails are easy to ignore. A prospect can always postpone an email, but on the phone, a prospect responds the moment he or she answers the phone. Email often falls into the multitasking category, but a phone call demands your attention. If you want a chance to actually sell, make a phone call first.

Emails are necessary in the prospecting phase. But phone calls are more important in the first contact. The interest you create on the prospect during the first call should encourage him/her reply your future emails and proceed with the prospecting process.

Sending an email to follow up with a phone call is a best practice. If you leave a series of voice mails, you can?t be sure your prospect is listening to them. If you speak to a prospect on the phone, a follow up email to thank them for their time and offer additional information can propel next steps, and gives prospects the option to contact you in the format they prefer.
